RESUMO

Recently, natural photosensitizers, such as berberine, curcumin, riboflavin, and emodin, have received more and more attention in photodynamic therapy. Tanshinone I (TanI) is extracted from a traditional Chinese herb Danshen, and exhibits many physiological functions including antitumor. TanI is a photoactive phytocompounds, but no work was tried to investigate its potential photodynamic effect. This study evaluated the cytotoxicity induced by the photodynamic effect of TanI. The photochemical reactions of TanI were firstly investigated by laser flash photolysis. Then breast cancer cell line MDA-MB-231 was chosen as a model and the photodynamic effect of TanI on cancer cell was evaluated by MTT assay and flow cytometry. The results showed that TanI could be photoexcited by its UV-Vis absorption light to produce 3TanI* which was quickly quenched by O2. MTT assay showed that the photodynamic effect of TanI resulted in more obvious inhibitive effect on cell survival and cell migration. Besides, the photodynamic effect of TanI could induce cell apoptosis and necrosis, lead to cell cycle arrest in G2, increase intracellular ROS, and decrease the cellular Δψm. It can be concluded that the photodynamic effect of TanI can obviously enhance the cytotoxicity of TanI on MDA-MB-231 cells in vitro, which indicated that TanI might serve as a natural photosensitizer.

After harvesting multiple costal cartilages, the local defect disrupts the integrity of the chest wall and may lead to obvious thoracic complications, such as local depression and asymmetry of the bilateral thoracic height. Decellularized materials have been used for tissue reconstruction in clinical surgeries. To apply xenogenic decellularized cartilage in costal cartilage defects, porcine-derived auricular and costal cartilage was tested for characterization, cytotoxicity, macrophage response, and tissue regeneration. Most of the DNA and α-Gal were effectively removed, and the collagen was well preserved after the decellularization process. The glycosaminoglycan (GAG) content decreased significantly compared to that in untreated cartilage. The decellularized auricular cartilage had a larger pore size, more pores, and a higher degradation rate than the decellularized costal cartilage. No apparent nuclei or structural damage was observed in the extracellular matrix. The decellularized auricular cartilage had a higher cell proliferation rate and more prominent immunomodulatory effect than the other groups. Two types of decellularized cartilage, particularly decellularized auricular cartilage, promoted the tissue regeneration in the cartilage defect area, combined with noticeable cartilage morphology and increased chondrogenic gene expression. In our research, the functional components and structure of the extracellular matrix were well preserved after the decellularization process. The decellularized cartilage had better biocompatibility and suitable microenvironment for tissue regeneration in the defect area, suggesting its potential application in cartilage repair during the surgery. STATEMENT OF SIGNIFICANCE: Autologous costal cartilage has been widely used in various surgeries, while the cartilage defects after the harvesting of multiple costal cartilages may cause localized chest wall deformities. Decellularized cartilage is an ideal material that could be produced in the factory and applied in surgeries. In this study, both decellularized costal cartilage and auricular cartilage preserved original structure, functional biocompatibility, immunosuppressive effects, and promoted tissue regeneration in the cartilage defect area.

Objective:To summarize the nursing care of a patient with antineutrophil cytoplasmic antibody associated vasculitis and autoimmune hemolytic anemia, hemophagocytic syndrome.Methods:A patient with antineutrophil cytoplasmic antibody associated vasculitis and autoimmune hemolytic anemia, hemophagocytic syndrome on March 24, 2020 in Jingmen Second People′s Hospital was selected. The key points in nursing were targeted nursing such as high fever, for complications(hypothermia and shock); actively monitor systemic damage and preventive care of complications; including prevention of exacerbation of hemolysis and prevention of complications after blood transfusion;observation and nursing of prevention of bleeding and disseminated intravascular coagulation;renal function monitoring, evaluation and prevention of complications; nursing of respiratory failure and prevention of complications,and medication nursing.Results:After careful treatment and care, the patient's body temperature returned to normal, and respiratory failure was corrected. There was no worsening of hemolysis, bleeding, disseminated intravascular coagulation, or renal failure. Hemoglobin, platelets, erythrocyte sedimentation rate, and serum ferritin were normal after repeated examinations, and the patient was discharged smoothly.Conclusions:According to the clinical characteristics and individual conditions of the patient,systemic damage monitoring and prevention of complications,accurate assessment and observation of the condition,and targeted protective measures are the keys to reducing systemic damage,complications and successful nursing care.

RESUMO

Objective:To assess the safety and efficacy of Anthocyanins for the treatment of Alzheimer's disease.Methods:From November 2018 to December 2020, a multicenter, double-blind, randomized controlled clinical study was conducted in 6 hospitals.The regular medication for the two groups was memantine, with the addition of a combination preparation containing Anthocyanins for the experimental group and a placebo for the control group.The Mini-Mental State Scale(MMSE), Montreal Cognitive Assessment Scale(MoCA), Alzheimer's Disease Assessment Scale-Cognitive Subscale(ADAS-cog), Activities of Daily Living Scale(ADCS-ADL)and Hamilton Depression Scale(HAMD)were used for assessment at the beginning.After 16 weeks of treatment, MMSE, MoCA, ADCS-ADL, ADAS-cog and the Clinician's Interview-Based Impression of Change Plus Caregiver Input(CIBIC-Plus)Scale were conducted and adverse events were recorded.Results:A total of 66 patients were enrolled, with 33 in the control group and 33 in the experimental group.There were no significant differences in cognitive function scores between the two groups before enrollment.Differences in MMSE scores, MOCA scores and ADAS-cog scores before and after treatment between the control group and the experimental group were 1.9±2.4 vs.3.4±2.0( t=2.62, P=0.011), 1.8±1.9 vs.2.9±1.4( t=2.45, P=0.018)and 3.0±2.3 vs.5.3±4.6( t=2.45, P=0.019), respectively.The differences were statistically significant.Instrumental activities of daily living(IADL)scores before and after treatment in the control group were 21.6±5.7 vs.22.6±6.2( t= 2.09, P= 0.046), and those in the experimental group were 22.7±5.4 vs.23.4±5.4( t= 2.45, P= 0.021). The differences between the two groups before and after treatment were statistically significant. Conclusions:Treatment with Anthocyanins can delay the decline of cognitive function and activities of daily living ability in patients with Alzheimer's disease.Anthocyanins may be a promising therapeutic drug for Alzheimer's disease in the future.

Objective To investigate the effects of standard large trauma craniotomy on neuron-specific enolase(NSE)inflammatory factors in patients withintracranial hematoma caused by severe trau-matic brain injury(sTBI).Methods A total of 64 cases of sTBI patients were randomly divided into con-ventional surgery group (conventional group)and standard large trauma craniotomy group (standard group),with 32 cases in each group.The postoperative Glasgow Outcome Scale(GOS)score,preoperative and postoperativeserum inflammatory factors(IL-8,IL-6,TNF-α,ICAM-1 ,and IL-1 0),NSE levels,and postoperative complications were compared.Results The GOS score of the standard group was signifi-cantly superior tothe conventional group(P 0.05 ).Conclusion Inflammatory factors and NSE both playimportant roles in sTBI.Standard large trauma craniotomycan control these indicators effectively and reduce the severity of the patient's illness.

Background and purpose: As a member of Catenin family, little is known about expressive signiifcance and mechanism of Delta-catenin in numerous tumors. In present study, we investigated the expression of Delta-catenin in breast cancer tissues and its correlation with the prognosis of patients. Methods:We examined the expression of Delta-catenin and its correlation with clinicopathological factors of patients by immunohistochemistry in 92 cases of breast cancer tissues with tissue micro-array. The mRNA and protein expression of Delta-catenin were also detected in another 32 cases of frozen paired breast cancer tissues and normal breast tissues. Results:Compared with normal breast tissues, the mRNA and protein expression of Delta-catenin were increased in breast cancer tissues, and expression of Delta-catenin was closely associated with histological grade and lymph node metastasis (P=0.016 and 0.022, respectively). Moreover, Kaplan-Meier survival analysis revealed that patients with high Delta-catenin expression had shorter survival than patients with low expression (P=0.015), and multivariate Cox analysis revealed that high Delta-catenin expression was also an independent prognostic factor (P=0.017). Conclusion:Our results suggest that Delta-catenin acts as an oncoprotein when overexpressed in breast cancer, and its expression is associated with poor prognosis of patients.

Objective To observe the clinical efficacy of cervus and cucumis polypeptide combined with Duhuo Jisheng Decoction Jiaji pain point injection in the treatment of lumbar intervertebral disc herniation. Methods Totally 68 cases of patients with lumbar intervertebral disc herniation were randomly divided into treatment group (34 cases) and control group (34 cases). Both groups were given Duhuo Jisheng Decoction orally besides basic treatment, and the treatment group received cervus and cucumis polypeptide Jiaji pain point injection additionally. Both groups were in a course of 10 days and treated for two courses. The clinical symptoms, visual analog pain scale (VAS), and Oswestry disability index (ODI) of the two groups were observed and compared among those before treatment, after one and two courses of treatment. Results Compared with the pretreatment of the same group, the VAS and ODI scores were lower after the first and second courses of treatment (P<0.05). After one and two course of treatment, VAS and ODI of the treatment group were lower than the control group (P<0.05, P<0.01). After one course of treatment, the total efficiency of treatment group was 91.2%(31/34), while the control group was 70.6%(24/34);after two courses, the total efficiency were respectively 100%(34/34) and 85.3%(29/34);the treatment group was better than control group during the same period (P<0.05). Conclusion Cervus and cucumis polypeptide combined with Duhuo Jisheng Decoction Jiaji pain point injection has good clinical efficacy in the treatment of lumbar intervertebral disc herniation.

OBJECTIVE:To study the influence of different herb composition on content of aconitine in Rhubarb and Aconite Decoction.METHODS:On the basis of Rhubarb and Aconite Decoction,different prescriptions were made up and different de?cocting technics were adopted,then the content of aconitine was determined with UV-spectrophotometry.RESULTS:The content of aconitine in mixture of decoctions of single herb decreased about20%and the content of aconitine in the decoction of combined herbs decreased about45%compared with that in decoction of single Aconite.Combination of Asarum with Aconite showed no influence on the content of aconitine.CONCLUSION:Rhubarb can control the toxicity of Aconite in Rhubarb and Aconite Decoction.

RESUMO

Objective To evaluate the safety of Fotemustine. Methods The studies had been done according to the experimental method of injection agents in pharmacodynamics guide principle on toxic test methods of application. The vascular stimulative assay of fotemustine injection on rabbit, the hemolyzation or agglutination of it in vitro, and the allergic reaction on guinea pig were performed. Results It was showed that the Fotemustine injection had no vascular irritation and injury effect on rabbit vascular endothelial cell. Compared with that of the control group, there was no difference. Conclusion No evidences of hemolyzation and aggregation on rabbit erythrocyte, and no allergic reaction on guinea pig were observed.
